Palbociclib is a targeted therapy for the treatment of hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced or metastatic breast cancer. It is not a chemotherapy drug but a kinase inhibitor that is always used in combination with other hormone therapies, such as an aromatase inhibitor or fulvestrant.

Palbociclib is used to treat adults with advanced or metastatic breast cancer. It is prescribed for postmenopausal women and men as an initial endocrine-based therapy. It is also used in patients whose disease has progressed after previous endocrine therapy.
Palbociclib is a type of medication called a kinase inhibitor. It works by blocking the action of specific proteins called cyclin-dependent kinases (CDK) 4 and 6. By blocking these proteins, Palbociclib stops cancer cells from dividing and multiplying uncontrollably.
Patients taking Palbociclib may experience side effects, which can vary. The most common side effects include:
Low levels of white blood cells (neutropenia), which can increase the risk of infection.
Fatigue or low energy levels.
Nausea, vomiting, and diarrhea.
Mouth sores or pain.
Hair thinning or hair loss.
Reduced appetite.
Seek immediate medical attention if you notice:
Severe or life-threatening inflammation of the lungs (pneumonitis), with symptoms such as trouble breathing, cough, or chest pain.
Blood clots in your lungs (pulmonary embolism), with symptoms like trouble breathing and chest pain.
Severe allergic reactions, which may include swelling under the skin, in the tongue, mouth, or throat, or trouble breathing.
Fever, chills, or other signs of infection, especially if you have a low white blood cell count.
Signs of low red blood cell counts (anemia), such as dizziness or shortness of breath.
Palbociclib capsules must be swallowed whole with food. Do not chew, crush, or open them.
If you vomit or miss a dose, do not take an extra dose to make up for it.
Avoid eating grapefruit or drinking grapefruit juice while taking this medication, as it can increase the amount of the drug in your blood.
Palbociclib is not for use during pregnancy, and effective birth control is required for both men and women during treatment and for a period after the final dose.
